This study enriched a mixed bio-cathode denitrifying culture (MBD) in a two-chamber MFC using repeated medium replacement without added organic carbon, achieving 69.99% NO₃⁻-N removal at 120 mV output. An isolated strain, Stutzerimonas stutzeri (Lyy), demonstrated >93% NO₃⁻-N removal across C/N ratios 2–12, pH 7–9, temperatures 20–40°C, and KNO₃ concentrations 500–3000 mg/L, with optimal complete denitrification (>98% TN removal) at C/N=4, pH 8, 35°C, and 2000 mg/L KNO₃. The work highlights electron shuttle-mediated electrotrophic denitrification and identifies Paracoccus and Pseudomonas as dominant functional genera.

Zentrale Erkenntnisse

  • Schwellenwert At C/N ratio 2, NO₃⁻-N removal reached 98.18% but NO₂⁻-N accumulated to 113.28 mg/L, yielding only 21% TN removal, whereas C/N ≥4 achieved >98% TN removal with no nitrite accumulation.

Was heißt das. Operators must maintain C/N ≥4 to prevent nitrite accumulation; carbon limitation forces preferential NO₃⁻ reduction over NO₂⁻ reduction, blocking complete denitrification.

  • Zielkonflikt Strain Lyy under anaerobic conditions achieved 98.53% TN removal in 60 h with 3.06 mg/L/h average rate, versus 22.42% TN removal and 0.92 mg/L/h under aerobic conditions—a 3.34× efficiency gain.

Was heißt das. Maintain strict anaerobic conditions in the cathode chamber; dissolved oxygen competes as electron acceptor and suppresses denitrification despite promoting cell growth.

  • Mechanismus At 40°C, NO₃⁻-N removal remained high at 100% but TN removal dropped to 20.43% due to NO₂⁻-N accumulation (OD₆₀₀ = 0.05), indicating nitrite reductase is more temperature-sensitive than nitrate reductase.

Was heißt das. Temperature control is critical; elevated temperature selectively inhibits the final denitrification step (NO₂⁻ → N₂), creating a bottleneck even when nitrate reduction proceeds.

  • Schwellenwert At KNO₃ 3000 mg/L, NO₃⁻-N removal was 97.05% but TN removal fell to 53.09% due to concentration-dependent NO₂⁻-N accumulation, whereas at ≤2000 mg/L complete TN removal occurred within 60 h.

Was heißt das. Limit initial nitrate concentration to ≤2000 mg/L KNO₃ to avoid nitrite accumulation; higher concentrations require longer residence times or staged treatment.

  • Methode The MBD enrichment method (six medium replacements over two batches) achieved 69.99% NO₃⁻-N removal in ~400 h, matching prior work (Ding et al.) that required fourteen replacements, demonstrating 2.3× faster enrichment.

Was heißt das. Use repeated medium replacement without organic carbon to rapidly enrich electrotrophic denitrifiers; this method outperforms gradual C/N reduction in speed.

Ansatzpunkte6

Konkrete Lücken in dieser Publikation, jeweils mit einem Experiment, das sie schließen würde.

  • Hoch No measurement of cathode potential (vs. SHE or SCE) during MBD operation; only voltage output (120 mV) reported. Optimal cathode potential window for electrotrophic denitrification is unknown.

Three-electrode characterization (Figure 2) was performed but only for electrochemical profiling; actual MFC operation used two-electrode setup with load resistance. Cathode potential directly controls electron availability and denitrifier community selection.

Versuch. Operate MFC with potentiostat control at cathode potentials ranging from −0.2 to −0.6 V (vs. SHE) and measure NO₃⁻-N removal, NO₂⁻-N accumulation, and microbial community shift.

  • Hoch No systematic study of HRT (hydraulic retention time) or current density effects on denitrification performance. Only batch experiments reported for strain Lyy.

MBD was tested in batch mode (Figure 1B shows discrete medium replacements); strain Lyy tested in 60 h batch cultures. Real wastewater treatment requires continuous or semi-continuous operation with defined HRT.

Versuch. Operate MFC cathode chamber in continuous mode at HRTs of 6, 12, 24, 48 h and measure NO₃⁻-N removal, NO₂⁻-N accumulation, and current density (mA/m²).

  • Mittel Electron shuttle identification incomplete. DPV shows a peak at 0.08 ± 0.05 V attributed to phenazines, but no chemical confirmation (HPLC, mass spectrometry) or quantification provided.

Text states 'it is speculated that the MBD possesses the ability to secrete electron shuttles, such as phenazines' but no direct evidence presented. Phenazine concentration and role in electron transfer kinetics unknown.

Versuch. Extract and quantify phenazines from MBD biofilm using HPLC-MS; correlate phenazine concentration with denitrification rate and electrochemical impedance.

  • Mittel No investigation of N₂O production or accumulation. Only NO₃⁻-N, NO₂⁻-N, and TN measured; N₂O not mentioned.

Denitrification pathway can produce N₂O as intermediate or end product, especially under low oxygen or high nitrite conditions. At C/N=2 with 113 mg/L NO₂⁻-N accumulation, N₂O risk is high but not assessed.

Versuch. Measure N₂O in headspace using gas chromatography during strain Lyy denitrification at C/N 2–4, pH 7–9, and elevated KNO₃ (2000–3000 mg/L).

  • Mittel Catholyte pH not controlled or reported during MFC operation. Only strain Lyy batch cultures tested at pH 4–9; MBD operation pH unknown.

pH is critical for denitrification kinetics and microbial community composition. Strain Lyy optimal pH is 8, but MBD enrichment and MFC operation may have drifted pH due to electrochemical reactions.

Versuch. Monitor and control catholyte pH during MFC operation at pH 7, 8, 9 using buffer or pH control; measure NO₃⁻-N removal, NO₂⁻-N accumulation, and microbial community.

  • Mittel No comparison of MBD performance with pure strain Lyy in the same MFC system. Isolated strain tested only in batch culture, not in bio-cathode MFC.

MBD achieved 69.99% NO₃⁻-N removal in MFC; strain Lyy achieved >98% in batch. Unknown whether strain Lyy alone would outperform or underperform the mixed culture in MFC, or whether synergistic interactions with Paracoccus and Pseudomonas are essential.

Versuch. Inoculate MFC bio-cathode with pure strain Lyy culture and compare voltage output, NO₃⁻-N removal, and NO₂⁻-N accumulation to MBD over 400 h.

Zusammenfassung

Recent advancements in microbial fuel cells (MFC) technology have significantly contributed to the development of bio-cathode denitrification as a promising method for eco-friendly wastewater treatment. This study utilized an efficient repeated replacement method to enrich a mixed bio-cathode denitrifying culture (MBD) within a bio-cathode MFC, achieving a stable maximum output voltage of 120 ± 5 mV and a NO 3 - -N removal efficiency of 69.99 ± 0.60%. The electrotrophic denitrification process appears to be facilitated by electron shuttles. Microbial community analysis revealed a predominance of Proteobacteria , with Paracoccus and Pseudomonas as functional genera. Additionally, the isolated strain Lyy (belonging to Stutzerimonas ) from MBD demonstrated exceptional denitrification efficiencies exceeding 98% when treating wastewater with a broad range of C/N (2-12) ratios and KNO 3 concentrations (500-3000 mg/L) within 60 h. These results demonstrated the effectiveness of the repeated replacement method in enriching bio-cathode denitrifiers and advancing MFC application in sustainable wastewater management.
